NAD+ stands for nicotinamide adenine dinucleotide. It is one of the most abundant and vital coenzyme central to many metabolic pathways, which declining levels associated with aging predisposes to mortality and morbidity. NAD+ is the coenzyme of sirtuins, which are conserved regulators of aging and longevity in diverse organisms. These enzymes have a central role in DNA repair, cell cycle control and mitochondria function. Dysfunction in these processes may also predispose in chronic aging related pathologies. For example, Disruption of NAD+ homeostasis due to mitochondrial dysfunction is central in the development of cardiac hypertrophy and heart failure. The metabolism of NAD+ also influences cellular senescence, which is stable growth arrest characterized by pro inflammatory cytokines implicated in tissue ageing and cancer. NR can improve many aspects of Alzheimer's disease neuropathology including neurogenesis, neuroinflammation, and cognition. Among other precursors of NAD+ including NAM, NA, and NR, only NR prevents axonal degenerations in neurons of mice. Axonal degeneration is one of the early events in chronic neurodegenerative diseases caused by excitotoxicity. This neuroprotective effect of NR to axonal degeneration may be caused by its ability to support sirtuin 3 and increase mitochondrial NAD+.
