According to Jack Dorsey, the biggest threat to Bitcoin is becoming irrelevant and not being used as a medium of exchange.
The Bitcoin White Paper reads "PEER TO PEER ELECTRONIC CASH" and some people are worried that it will not fulfill it's original vision.
Bitcoin on layer one could never be used as a medium of exchange.
However, with new tools on layer two (lightning, liquid, ecash), it is definitely capable of becoming a medium of exchange as well.
