Jacob and Evie were sent by George Westhouse to kill the Templars Rupert Ferris and David Brewster, respectively.

As Jacob and Evie arrived in London, they sought out the Assassin Henry Green to devise a plan to take down the Templars in the city. Jacob and Evie met with Sergeant Frederick Abberline to gain allies in the Metropolitan Police Service. Jacob and Evie met with Clara O'Dea. To secure an alliance with her, they agreed to free several child workers from a factory. Jacob and Evie set out to kill Rexford Kaylock and take over Whitechapel once and for all.

Henry took Evie and Jacob to meet Alexander Graham Bell, a scientist and inventor who would be able to repair the rope launcher taken from Rexford Kaylock.

The Secrets of London are collection of music boxes scattered throughout London by the Assassin Michel Reuge; inside the music boxes were discs, marked with the Assassin insignia, that acted as keys for Reuge's Vault, in which he presumably hid a Piece of Eden. Jacob and Evie helped Alexander Graham Bell recover the cables he had ordered for his independent telegraph line.

While Jacob investigated Starrick's Soothing Syrup, Evie followed a lead on the Piece of Eden. Jacob investigated the source of Starrick's Soothing Syrup. Jacob searched for the creator of Starrick's Soothing Syrup by investigating its distillery. Jacob questioned Richard Owen to learn who was behind the production of Starrick's Soothing Syrup.

While Jacob investigated Starrick's Soothing Syrup, Evie followed a lead on the Piece of Eden. Evie and Henry infiltrated the Kenway Mansion to continue the search for the Piece of Eden.

Having learned that John Elliotson was behind the production of Starrick's Soothing Syrup, Jacob set out to assassinate him.

Jacob and Evie once more met with Alexander Graham Bell again, who had produced an incapacitating bomb for them. While Jacob worked on ending Starrick's control of London's transport, Evie searched for the Shroud of Eden at the Monument.

In order to end Starrick's monopoly on London transport, Jacob sought out his main competitor, Pearl Attaway. Pearl Attaway tasked Jacob with stealing a set of internal combustion engines from the Millner Company.

With the internal combustion engines in her possession, Pearl Attaway charged Jacob with assassinating Malcolm Millner. Having learned that Pearl Attaway would be at Waterloo station, Jacob set out to assassinate her.

Charles Dickens welcomes the Frye twins as members of the "Ghost Club", and together they investigated local mysteries with supposed paranormal causes.

The Frye twins visited Charles Darwin in the Strand.

Evie visited Southwark to see how the borough was faring after Jacob had killed Pearl Attaway. Evie infiltrated the Tower of London to find the Shroud of Eden.

While Evie went after Lucy Thorne, Jacob learned that a banker known as "Plutus" was working for Starrick and set out to discover his identity. Jacob trailed the Cockham Merchants weapons shipment to its destination to uncover the identity of "Plutus". Jacob set out to assassinate Philip Twopenny before the latter could successfully rob the Bank of England.

After the death of Philip Twopenny, Britain's economy was left in danger and counterfeiters had stolen printing plates from the Bank of England. Sergeant Abberline thus had Evie recover the plates to rescue the British economy. Evie and Henry set out to steal the plans for the vault holding the Shroud.

While Evie continued her search for the Shroud, Jacob had discovered a Templar plot to assassinate Prime Minister Benjamin Disraeli, and set out to discover the identity of the man behind it, who was only known as "B". Jacob set out to rescue Prime Minister Disraeli and learn from him the identity of "B". Jacob escorted Mary Anne Disraeli through the Devil's Acre in exchange for information in the identity of "B". Jacob set out to assassinate the Earl of Cardigan at the Palace of Westminster.

Jacob received an invitation to meet with Maxwell Roth, the leader of the Blighters, who proposed an alliance against Crawford Starrick. Jacob helped Maxwell Roth kidnap three of Crawford Starrick's important associates.

Jacob and Maxwell Roth set out to destroy one of Crawford Starrick's workshops. Jacob resolved to assassinate Roth, who had invited him to a performance at the Alhambra Music Hall.

After disagreeing about the former's methods, Jacob and Evie decided to kill Starrick and recover the Shroud in one final mission together. To do so, Jacob would need to acquire invitations to a ball at Buckingham Palace. Jacob needed to acquire a Royal Guard uniform for Sergeant Abberline, who could then smuggle in weapons for the former and Evie at the Buckingham Palace ball. Evie drove for Duleep Singh and several politicians in exchange for information on the whereabouts of the plans to the Buckingham Palace vault.

Jacob and Evie infiltrate the Queen's Ball at Buckingham Palace to confront Crawford Starrick and secure the Shroud of Eden.

Jacob and Evie are tasked by Queen Victoria to stop a dangerous plot led by a surviving faction of Templars. Queen Victoria tasked Jacob or Evie with learning the motives of the captured Templar leader. Queen Victoria tasked Jacob or Evie with recapturing the escaped Templar leader. Queen Victoria charged Jacob or Evie with stopping the Templars from detonating the House of Parliament.

The Initiate approached a rift in the Helix simulation, beckoned by a voice.

Lydia went to speak with Winston Churchill in the Tower Bridge regarding the spymaster they were hunting.

Jack the Ripper and Jacob Frye face off against each other, each determined to rid Whitechapel of the other. Evie met with Inspector Frederick Abberline, who brought her up to speed on Whitechapel's situation and then escorted her to the first crime scene. Evie set out to find clues in Jacob's residence, hoping to find some clue as to the location, or identity, of Jack the Ripper. Evie traveled to Owers Manor outside of London to assassinate Olwyn Owers, the lieutenant of Jack the Ripper.

Evie met with Inspector Frederick Abberline of the Metropolitan Police Service at Scotland Yard, hoping to gain information on the whereabouts of Arthur Weaversbrook before Jack the Ripper could get to him. Evie sailed downriver to Deptford in order to question and free Jack the Ripper's hostages on the prison hulk ships and in the abandoned garrison. With his plans in Deptford foiled by Evie Frye and the Metropolitan Police Service alerted, Jack the Ripper sought to eliminate any evidence and witnesses that may have compromised the Assassins.

Having returned from Deptford, Evie convened with Inspector Frederick Abberline and the Metropolitan Police Service at the murder scene of Mary Kelly.

In order to erase his identity completely, Jack returned to Lambeth Asylum to eliminate any medical staff with memory of him and destroy all records of his internment there. Evie made her way to Lambeth Asylum in order to kill Jack the Ripper.
